motherboard pci- questions

https://www.msi.com/Motherboard/MAG-X670E-TOMAHAWK-WIFI
I have this motherboard listed above. my question is simply, if i put a pci x 2 card in the bottom most pci - e 16x port, would it stop my graphics card operating at pci-e 16?

No, if your graphics card is in the 16 x slot then that is what it will operate at.
