The benefit of say Comodo over a router firewall is that the router tends to only stop incoming connections from the internet. If you got infected with a virus, either by visiting an infected website or other means, the virus tends to make a connection out from your computer to a control centre or whatever on the internet so a router won't stop that from happening. Software firewalls like Comodo will detect a lot of outbound connections by programs so adds another later of protection.
