The most common error during the launch of iOS5 was related to Apple's servers being hammered. Have you tried since then?
If you already have iOS5 downloaded...
Just try "Update" within iTunes. It shouldn't be a problem now.
If it is a problem, or it tries to spend ages performing a backup, then instead of clicking on Update, shift-click on Restore and browse to the firmware location (e.g. on Windows, it's "C:\Users\<name>\AppData\Roaming\Apple Computer\iTunes\<iDevice Name> Software Updates\*.ipsw"
